=B7 To the best of my knowledge I have not seen any longitudinal data tha=

validated the number of infected cells with clinical outcome. We have do=

something a little bit different than what was originally published by =
Schmittman at the NIH when attempting to quantitate the number of infecte=
d cells =
in HIV. He used limiting dilution's followed by PCR for the gag products=
=2E In =
our approach we sorted cells into dilution's starting at 10K and ending a=
t 10 =
cells followed by PCR for the gag products. These were measured by a tec=
hnique =
known as the liquid hybridization protection assay which is a qualitative=
assay =
for the presence of these products. These results were only studied on a=
cross-
sectional basis and were only intended to answer questions related to oth=
er =
infected cell types such as monocytes. However, I wide range of results=
were =
seen, some positive at <100 cells and other still negative at 10,000 cel=
ls. =
This might lead one to believe a correlation might exist with clinical ou=
tcome.

The clinicians at our center has confronted me with a few relatively =
simple questions, which I am not able to answer and therefore has =
forwarded to the List of Flow'ers:
We have been wondering if anybody has used flow cytometry to count the =
number of infected cells in HIV-1 patients. Any communications of =
correlation to molecular biology (ie. semiquantitative HIV-1 RNA-PCR)?
If so, is the infected fraction predictive of clinical outcome of HIV-1 =
infected patients? =
