When I worked at BD, I tested the AO protocols on a FACS 440 and after
increasing the AO concentration, the dye worked like a charm.
Cleaning the instrument requires no more than running diluted house hold
bleach for 10 minutes. Do make sure you fill the sample tube, so that all
AO on the outside of the sample tubing is removed. Verification of the
complete removal of AO can indeed be done with control samples, however,
make sure you have rinsed out all the bleach first, otherwise the reading
is meaningless.
Used at high concentrations dyes like Hoechst, Propidium Iodide, Ethidium
Bromide, LDS 751, Thiazole Orange are just as 'unpleasant', so you may want
to adopt a bleach rinse as a routine procedure between users.
