Image Analysis Systems?

A close colleague at a nearby lab has asked me to post the following question.
Replies can be either sent directly to me or posted on the net if you think
that the information is of general interest. Actual reviews of specific
packages both good and bad would be most appreciated.

I am in charge of a histology/ cytometry laboratory in Gaithersburg, MD. We
are doing immunoperoxidase/alk. phos. staining for various antigens in tissue,
cytospins, or cells cultured on coverslips or slides ( cell preps are handled
in this way when flow cytometry cannot be done) . I am looking for a good
image analysis system that will enable me to quantitate positive-staining
subpopulations in cell preps , or determine the proportion of positive-
staining cells in a defined area in tissue (i.e. PCNA-positive cells in
bronchial epithelium). Automated stage movement would be needed and slide
autoloading would be a welcome capability as well. Since we have also been
obtaining good multi-level staining for some antigens, it might be useful to
have some densitometry capabilities in the software, although this may be
considered icing on the cake or a much more sophisticated function that may
not be possible with one software package. Color discrimination is preferred
over gray-scale discrimination. To complicate matters even more, I would also
like to be able to use the same hardware to handle images from a conventional
mercury lamp fluorescence microscope, although at this time I am placing
priority on light microscopy, since that is where our quantitation needs are
the most immediate. I have a Nikon Optiphot with Nomarsky capabilities that
I use for light microscopy, and a Nikon Labophot which I use for my
fluorescence work. Is there anyone out there who can direct me to a few good
contacts so that I can put together a system that will work for me? Thank you
for your help!
