Hi
I have used Hoechst 33342 to DNA stain viable cells and sort on grounds
of their DNA content for some time. It works pretty well. Usually 10
micrograms per ml works pretty well although you may have to play about
with concentration, much higher than this and the cells do not grow well
if at all. I stain for at least 30 mins at 37 degrees by just chucking it
in with the cell culture. I keep then on ice prior to staining.
