Fixation for 2-colour: apoptosis + surface antigens...

Kruger Gray asked about fixation and staining. Both formaldehyde
and ethanol will alter surface antigens so that they do not react
with their antibodies. Therefore, the antibody reaction must be
performed before the fixation. Then fix with formaldehyde.
The formaldehyde will permeabilize the cells, but treatment
with 0.1% Triton does a more complete job. After fixation you can
stain for PI.
