Bruce, We have been working in collaboration with some
cytogeneticists to look at foetal reticulocytes in maternal blood,
purifying them for genetic analysis. The frequency of adult reticulocytes
seems to be extremely low in healthy women, but foetal cells are
relatively frequen, about 1/10,000 of nucleated blood cells. This
still means a lot of cells on a cytometry plot to get a meaningful
answer. We are using magnetic beads to pull them out using the
simple equation of CD45-negative, CD71-positive. This operates on
the principle that they are nucleated cells, not leucocytes, but express very high
levels of the transferrin receptor to bring in iron for haemaglobin
synthesis.
