In response to Eric Miller's question about a backup method for Annexin V,
the lipophilic dye merocyanine 540 (MC540) binds to cell membrane
phospholipids when they unpack early on in apoptosis. This phospholipid
unpacking is analogous to the loss of membrane asymmetry and the
subsequent exposure of phosphatidyl serine that Annexin V detects.
A group of investigators here have been using MC540 correlated with
propidium iodide uptake as an early indicator of apoptosis. There
are other investigators out there also using this dye to study
apoptosis correlated with other markers. For more info I would
suggest reading a paper published last year in the Journal of
Immunology => Decreased Membrane Phospholipid Packing and Decreased
Cell Size Precede DNA Cleavage in Mature Mouse B Cell Apoptosis,
Mower et al, Journal of Immunology, 1994, 152:4832.
