Cell surface fluorescence after Fix & Perm

Hi Everyone!

We have experienced a dramatic decrease in PE fluorescence using CD4-PE
CD1-PE and CD45RA-PE after treating the cells with Fix and Perm from
Caltag when we stained human thymocytes and a whole blood preparation of
human PBL. Other antibodies conjugated to PE show only minor flurescence
decrease. In addition the same clone of CD4-PE maintaned its
fluorescence on PBL, but not on thynocytes.

Has anybody experienced similar problems? Can you suggest a solution on
how to prevent this? So far Caltag hasn't been able to provide anithing
but simpathy...
