Green laser and Cy3

Hello,

We would like to run Cy3 stained cells on our FACStar+ cytometer. We are
looking at the low number of binding sites and as a result of that we might
end up with only 3K-20K of Cy3 molecules per cell present. Excitation peak
for the Cy3 is ~550 nm with a significant shoulder starting at ~514. So it
seems like one can have a choice between a very week line close to
excitation maximum coming from a HeNeo laser (544 with 1-2mW) and a
stronger line from YAG or Argon lasers(50-400mW) which is suboptimal for Cy3
excitation.

Is there anybody out there who has some experience using Cy3 with agreen
laser on a FACStar+ ?

Any information would be very much appreciated.
