HLA-B27 / CD16a vs CD16b / TNF-production / Mac Problems

Hello to all with several questions:

1. We are currently discussing to
start with a specific HLA-B27 assay
by flow cytometry or alternatively PCR
as a supplemetary test to
a conventional cytotoxicity based
assay. Regarding flow cytometry I am
aware of 4 companies who all claim
that only their specific clone is charge
independent stable in conjugation and
selective against HLA-B7.
Any specific experience?

2.In a research project we are interested
in the specific analysis of either CD16a
(the transmembrane NK and monocyte
type) or CD16b (the neutrophil antigen).
I could not find a commercial source for
CD16a vs CD16b specific antibodies
(e.g. 1D3).
Any help?

3. Has anyone experience with a high
affinity antibody against TNF-alpha for
intracellular analysis of cytokine
production in monocytes?

4. We regularly encounter file structure
problems on the harddisks of several
Mac Quadras 650 linked to a couple of
HP340s, MS-Windows PCs and
Sun Sparcs in our local flow net. The
typical error on the machines only running
cytometry software from BD is an error
in the header node in the catalog b-tree
leading to large amounts of file garbage.
Is this problem related to the network
configuration?
