In response to Eric Miller's query about calcium on a FACScan:

Either of the free packages WinMDI (Windows) or MFI (PC) will plot
a parameter vs. time. MFI has some additional kinetics capabilities,
including calculating a ratio, and plotting the median value vs. time
or vs. event number (if your acquisition software doesn't acquire time
as a parameter). You'd need to move your listmode data from the HP
to a PC. My catalog of free flow cytometry software has info on the
options available for doing this:
