Dr. Watkins inquired about the simple AO procedure of cell staining
for DNA and RNA. The methodology of use of AO for differential
staining of RNA vs. DNA is presented in detail in Vol 41 of Methods
in Cell Biology (1994; Chapter 26, pp 401-420). Once the solutions
are made the procedure is rapid - it takes only 2 minutes from taking
the cell suspension directly from the cell culture to measuring its
fluorescence. The most essential requirement is the proper
concentration of AO in the staining solution.
