A negative cell control is not necessary for FCM TdT, but the use of the proper isotype
control is critical! If you are using one of the multiple monoclonal preps, like the one from
Supertechs, you need to mix the appropriate isotype controls in the same proportions.
Apparently, the mixed anti-TdT is stable but the mixed isotype controls are not. I've tried
keeping them after mixing, and there is a considerable increase in nonspecific staining,
making the mixture useless as an isotype control. The single monoclonal prep from
Supertechs has failed to react with some ALL's, at least in my experience. Too bad; it's
much cleaner. Just as an aside: a useful combo for detecting ALL is TdT/Cd19/CD10; very
pretty when you have a classic case.
