Actually, if you sort CD34+ cells you will not find CFU in the "dim"
population. Some of the early controversy over whether CD34+ cell number
could predict engraftment following stem cell transplantation or even
whether CD34+ cell number predicted CFU-GM content in a bone marrow graft
resulted from some investigators who included this "dim" population-which
clearly does not correlate with either of these parameters. Of course it
is true that if you put CD34+ cells into culture with growth factors,
eventually they lose CD34 expression, and at some point will be dim---The
point is if you want to study CD34+ cells-that is characterize them, or
use the information for evaluating stem cell products, I would not
include this dim population-I think I am in the majority of the CD34
community in this regard.
