ANTI-THROMBOCYTE ANTIBODIES IN ITP

WE HAVE BEEN LOOKING INTO THE POSSIBILITY OF USING FLOW CYTOMETRY TO DETECT
THROMBOCYTE BOUND IG ON PATIENT THROMBOCYTES AND THE BINDING OF PATIENT
SERUM IG TO CONTROL THROMBOCYTES BY FLOW IN ITP.
WE FOUND, ESPECIALLY IN THE DIRECT STAINING OF THROMBOCYTES, QUITE A GREAT
VARIABILITY ALSO IN "NORMAL CONTROLS".
WE ARE USING FRESH THROMBOCYTES ENRICHED BY CENTRIFUGATION AND ARE STAINING
LOG SCATTER-GATED THROMBOCYTES IN SINGEL COLOR ANALYSIS FOR HUMAN IG-G AND
IG-M.

I FIND OUR RESLUTS "LESS THAN OPTIMAL!!"

IS THERE ANYONE WHO BELIVES THAT FLOW CYTOMETRY IS A CLINICALLY USEFUL
METHOD FOR THIS ANALYSIS IN ITP??

ARE THERE ANY GOOD PROTOCOLS AVAILABLE AND IS THERE ANY FOLLOW UP AS TO THE
SENSITIVITY AND SPECIFICITY OF THIS OR OTHER THROMBOCYTE TESTS IN ITP??
