Dexamethasone is not a good way to induce apoptosis in peripheral
human T cells. Our standard control for many assays is very simple:
Mouse CTLL2 cells are readily available and are very highly dependant
on IL-2. 24 hours before you wish to do the experiment wash some of
these cells and put them back into culture with IL-2 (negative
control) or without IL-2 (positive control). This is extremely
reliable, but you may need to ficoll the cells first if you don't
split them often enough, or your cultures will all contain apoptotic
cells.
