Fish Lymphocytes:DNA Histograms

Thanks to all regarding how to isolate fish lymphocytes. The old tried and
true "experiment with diluting ficoll hypaque" worked out after all! Now
here is the other most interesting problem. Before I was able to isolate
the lymphocytes, I took a look at the "buffy coat" by staining with P.I. and
looking at the resulting DNA histograms. I got something quite interesting.
The RBC's which are nucleated in the fish, gave what I considered to be a
"normal" G1-S-G2/M histogram pattern--but further on down the fluorescence
scale--I saw another G1-S-G2/M pattern. Is this indicative of WBC's in the
fish i.e. does the DNA of WBC's stain more intense than the DNA of the
RBC's?

Help!!
