Potassium and Membrane Potential; Bead Quenching

Heinz Eipel asked about potassium probes and/or membrane potential dyes.
In most mammalian cells, potassium concentration gradients are a major
factor in membrane potential, and the standard cyanine (DiOC5(3), etc.)
or oxonol dyes should reflect changes in transport through channels.
In responding to a query about quenching beads, Alica Givan mentioned
Trypan blue. The problem with beads is that if the dye is dispersed
throughout the bead, it will be difficult to quench it because water
soluble quenchers such as Trypan blue won't get through the plastic,
and it's not likely that hydrophobic quenchers and solvents could be
applied at concentrations which wouldn't screw up the specimen.
