Mac antibodies responses

To all:

Happy Twelfth Night! On December 22 I posted a request for suggestions for an antibody
which could be used to quantitate mouse spleen antibodies. I thank all those who
responded. Following is a collection of the responses.

from Dennis Broud:

"PharMingen's catalog # 0178sB, Anti Mac-3...PE labelled Rat IgG1 product..."

- From Paul K. Wallace:

"F4/80 is the best if the mp are not activated. Mac-1 (CR-1, CD11b) is good but is
also on B-cells and granulocytes. Mac is only on activated mp ....from the ATCC &
Serotec....Use a murine Ig (Sigma) to pre-block, mouse serum works but it is not as
good. A goat antirat F(ab')2 conjugated with PE (caltag) will work better that FITC
due to mp autofluorescence."

."Mac-1 is specific for activated macrophages, while Mac-3 is specific for
macrophages and neutrophils....from ATCC"

"...anti-mouse Mac-1..." but it also "supposedly picks up Ly-1 B cells..."

"...(Mac1) from either Caltag of PharMingen. The trick is to use directly conjugated
pe or pe/cy5-av with biotinylated antibody so the the autofluorescence is lower. We
also preincubate with Fc block, a rat anti mouse FcR FAB fragment which blocks the Fc
receptors on the macrophage cell surface (available from PharMingen or from ATCC as
hybridoma 2.4g2 I think..."
