Na+ and K+ measurements

Christopher Worth asked about experience with sodium and potassium probes.
We have used SBFI with image analysis and Sodium Green with flow and get
satisfactory results. We ratio sodium green fluorescence on forward angle
light scatter to partially correct for cell size, since it is not a dye that
has a wavelength shift upon binding to sodium. Nevertheless, we have
measured both baseline levels in different cells and also modulated sodium
levels with drugs and everything seems to work properly. We presented a
poster at ISAC and you can get some more information from the abstract.

There are no good potassium indicators that I am aware of, however. PBFI
simply does not work because it has very little change in intensity or
spectrum shift with binding to potassium. The only alternative is to measure
membrane potential with oxonol dyes, for example, and infer potassium changes
from that.
