We have used TO-PRO-3 iodide (TP3) as an alternative for
propidium iodide in order to measure DNA content. In this way it
is possible to combine DNA measurements with the analysis of
the binding of two antibodies against cytoplasmic or nuclear
antigens (indirect immunofluorescence with FITC and PE). These
experiments were performed on an Elite with 488nm excitation for
FITC and PE and 633nm excitation for TP3. The technique is
described in the last issue of Cytometry (17:185-189, 1994).
