Volume measurements

We are doing a lot of glutathione measurements in leukemia samples, and
are attempting to calibrate these to give GSH content as fmol/cell. Since
GSH-based detoxification processes are enzymatic reactions, I think it
would be more appropriate to express results as concentration. To do this
I need a measure of cell volume. Forward light scatter seems a bit tacky,
and I would need to spend money getting my old FACS Analyzer up and
running, although this would otherwise be a good solution as it has a
Coulter orifice.
I seem to recall that there are methods for measuring surface area using
dyes that partition into the outer leaflet of the plasma membrane, which
would then allow derivation of volume, and I would be very interested in
hearing from anybody with experience in this field.
