Hello Sania,

 

Did you examine by microscopy whether bacterial DNA (in the bacteria invading your cells) is not TUNEL-positive? It is possible that being degraded bacterial DNA may become a good primer to terminal transferase and undergoes labeling during the TUNEL reaction. Besides, microscopy is the only sure way to distinguish apoptosis from necrosis.

Using the Tunel staining I've obtaining higher values in comparison to negative control cells (30%). Using Annexin staining the values are very low, about 1-5% of apoptotic cells. Otherwise the PI staining indicate necrosis in about 20% of cells. How can I interprete these results?
