The samples I will recieve will have differing DNA content, which I need to use to put a number to each cell type. The method suggested was with Hoecht 33342, but we do not have a UV laser. The methods I have cobbled together are 1) Fix and stain with PI, or 2) Stain with syto-9 (part of Invitrogen's Live/Dead discrimination kit. I just wondered if anyone did this sort of thing routinely, and if they had a protocol I might try.
