Would anyone care to comment on incompatibilities that may arise between PKH staining and surface immunostaining for flow (other than usual conflicts in fluorochrome selection). For example does PKH have any affects on subsequent antibody binding and therefore modify antibody concentrations or incubation times/temp, etc? Related to this, if anyone has done concurrent PKH staining and immunostaining of frozen tissues (with or without a permeablization step) I would also welcome advice. Thanks so much!
