Hello,



Here is a brief summary of the answers I received concerning the antibody
conjugation methods.



Several people that answered used the Molecular Probes Alexa kits for 100µg
of antibody with success. The Qdots labelling seems to work as well.



Concerning the Zenon conjugation that allows the staining of the exact
quantity of antibody needed for one tube/experiment, I received divergent
experiences. This staining seems to be simple and rapid however one
mentioned that "the complexes dissociate rather rapidly (for some
antibodies) and have to be used within 30-60 minutes of forming the
complexes for reasonable performance". Notably, it seems that the
conjugation is dependant of the antibody heavy chain type, thus you have to
choose carefully your kit depending on your antibody. In conclusion, it
seems to work, is expensive for a systematic use but may be useful to test
new antibodies/fluorochrome combinations.



Another product from Innova Biosciences has been mentioned twice as a good
alternative, notably the conjugation seems stable up to one year.



Nobody had experience with the kit form AbD Serotec.



Finally we choose to get one Alexa microscale kit from MP and one Zenon kit
to try and to use with exotic antibodies.
