I suggest you to check previous answers in the list during this year. There was one from me plus many others from different colleagues. The main problem is that you cannot use just one marker to differentiate memory subsets in the PB or tissues, since CD4+ T cells are extremely heterogeneous. Moreover, many of these markers, if not all, are affected by activation in vitro since activation itself generates effector cells (for this check Sallusto et al., Ann Rev Immunol, 2004).
