Hi B., safest is always to trash your contaminated cells and start up from a new cyrovial - if you have that luxury. We ourselves sometimes receive contaminated transfectants/hybridoma's/weird and hard to get cell lines, so there's no chance of going back to a safe in-house stock. In those cases, I treat them at least one week with MRA (from ICN, now MP Bio), then serially with BM cyclin 1 and 2 (from Roche), as instructed by the supplier. The triple-whammy usually does the trick, as monitored by re-emergence of mycoplasma contamination after all treatment is withdrawn for at least one week after the last round. Of course, you should always keep those cells in a separate incubator (where facilities allow), not handle them simultaneous with uncontaminated cells in the same hood,... untill you're really sure they're clean. We have a special "quarantine" incubator for newly arriving cells, which is also capable of sterilizing itself afterwards.
