Dear Flowers,

I need your help to figure out what's going on with my 6 color compensation
on FACSCanto.
As you can see at the plots (word document attached), I have problems to
compensate APC x PE-Cy5, is it normal the overlap be that high? I have to
compensate more than 70.
This is an example of human PBMC labeled with CD44 FITC, CD8 PE, CD4 APC,
CD62L PE-Cy5, CD25 PE-Cy7, CD69 APC-Cy7. But almost all my 6 color tubes
have the same problem, doesn't matter which antibody I am using.
I don't know if there is any correlation, but with some cells, especially
DCs, the isotype is always positive for APC and PE-Cy5.
Sincerely, I don't know what to do, any help will be really appreciate.
Thanks again
Ana.
