quality controls for CD34 enumeration (ISHAGE) on BD FACSCALIBUR

Hi!

I would like to know if someone has experience with BD Stem Control kit?
Our lab used to run STRECK controls for CD34 using ISHAGE protocols and
they have been reliable. Now, we're testing BD controls in parallel with
Streck's and the performance is really different between them. For BD
controls, we can't get enough events for CD 34 positive ( it is supposed
to be 100CD 34 events at least) and the data looks with more debris even
we use an exclusion gate for that purpose. The acquisition rate is very
low so the instrument can't reach the total events. We need to stop the
run at some point and save the date before the sample is gone. The
instrument is set up to acquire 100,000 total events.
Besides this if someone is running BD controls with ISHAGE on
FACSCCALIBUR we would like to discuss protocols to see details that we
might be missing.
We'll appreciate some collaboration on this topic.
Thanks a lot!
