Hi Flowers,

I am new to DNA analysis by flow so please bear with me.

Can anyone tell me how I can tell the difference between 2N cells that are in G2/M phase and 4N cells in G0?  On my gated histogram they fall on top of each other, correct?  With my positive control, I can see a very small 8N peak that tells me that I have hyperploid cells.  In my experimental condition, however, I am unable to say if the peak I see is only 2N cells undergoing regular cycling in G2/M, or if it is a G0/G1 peak of 4N cells.  I’m guessing that the only way to distinguish this is to look for additional peaks representing 4N cells in S or G2/M (or 8N peaks, etc) but thought I would see if there’s anything I’m missing.

Thanks,
Kerry
