Hi Paolo, I have tried to detect side population on our FACS Aria (with violet laser not UV) with Hoechst and the excitation was minimal. However, I was able to see SP in mouse bone marrow and some cancer cell lines using DyeCycle violet (from Invitrogen). However, the resolution was not as good as FACS Vantage when compared in parallel using an UV source. I also heard that the UV laser on FACS Aria is not powerful enough for SP detection but I have not validated that myself. Just my 2c. Italian BD sales offices say that a custom FACSAria with UV laser could be cost up to 400K Euro if we replace violet standard instead around 350K Euro for standard configuration. My question is : is it possible identify Hoechst SP by Violet or is it difficult to use? Additional question is: is it possible and is it easy use FACSAria as only analyzer instead to sorting?
