Dear FLOWers,
Thank you all for your prompt response regarding my previous flow cytometry queries. I
have one more now.
We have obtained a K562 cell line lot from NCCS, Pune, India, maintained in RPMI 1640
(NCCS suggested IMDM, but said RPMI also is O.K) and used for human NK cell cytotoxicity
experiments, using DiOC18 & PI flow cytometry. We find that the cytotoxicity is low, i.e
~10% in many healthy subjects, even when we use PBMC: K562 target 50:1. The figures
quoted in literature are usually much higher at this ratio.
We suspected the cell line and tested; we found that the cells are not expressing CD45,
as to be expected from reported literature.
If anyone has got a good working K562 cell line, could they a share some with us?
I would also invite any other suggestions for making our existing cell line work
better. Or are there any other cell lines for this purpose better than K562?
  Sincerely,
  Alamelu Raja,
  Tuberculosis Research Centre, Chennai,
  India.
