Hello fellow flowers, I have an investigator that is wanting to use flow cytometry to measure the number of mitochondria in plant protoplasts using a mitochondrial specific dye. He will have a small amount of cells (<1000) and wants to be able to identify number of mitochondria in each cell. I feel this is outside of the range of our instrument (BD FACSCAnto) and probably flow cytometry all together, but he has asked for me to get a second opinion. He also would like to measure relative amounts of mitochondrial DNA from isolated mitochondria. Does anyone out there have any experience with staining and measuring DNA from isolated mitochodria? If anyone out there has any advice, experience, or general discontent referring to this experiment please feel free to help me out. Also if anyone has any ideas as to how best to work out this experiment on any other instrument as well.
