Dear fellow flow-ers: I am trying to sort some cells for a client and am having real trouble. The cells are porcine intestinal epithelial non-transformed primary cell line isolated from the jejunum. They are similar to CACO-2 cells. They are 30-40 um in size. I am trying to sort them on a MoFLo sorter using a 150um tip. They appear to be especially susceptible to destruction and rupture. We have tried fixing them but they still seem to rupture and splouge(their word for spilling all the cell contents into the solution). I was under the impression that once fixed, the cells should stay stable and not rupture. Any help would be greatly appreciated. Thanks as always.
