Hi Mark,

I can sympathize - but let's be creative: I've used irrelevant target
specific humanized mAbs (say, cancer specific one on freshly isolated
leucocytes) as negative controls for isotype matching humanized ones
that binds surface markers (surface receptors, CD's, ...). You could
also think about using a soluble protein specific one as a negative
control for a surface specific one (say, anti-TNF, but be careful with
that one - there's documentation for surface expression for some of this
type of targets). You can get most of these from your local pharmacist,
as there are plenty of humanized monoclonals in the clinic already -
just try to find a doctor to write you a script where needed, or beg for
leftovers at your local hospital pharmacist (many are administered in
the hospital as infusions. Once reconsituted/thawed, most of these drugs
cannot be used later on, as per product insert administration
procedures. Since most to all are dosed according to the patient's body
mass, not as a fixed amount, there's bound to be some stuff left over in
the vial - which is good enough for science if kept at 4C, but certainly
not for use as an i.v. drug).

Alternatively, and very accessible/cheap to boot, you could just
purchase some human plasma isolated polyclonal IgG1 from Sigma!

have fun,

Guy

Subject: Control Abs

Hello, again,

As I said in the earlier post, I have a humanized antibody for use in
flow. Can I use an antibody(ies) raised in mice, or unicorns for that
matter, as isotype controls?????

I am having trouble (as you might expect) finding human abs.

He'p me, please.
