200 micron nozzle tips

Dear Flow-ers,

I'm about to embark on a sorting project that includes using a 200 micron
nozzle tip on my MoFlo. My experience up until now is based entirely in the
world of 70 micron tips. I have Dako's CytoCalc and a few personal
recommendations for tips that are bigger than 70 but still smaller than 200.
Would anyone working with larger tip sizes (ideally 200 micron!) be generous
enough to share any info they have about this process?	Mainly what pressure
you operate at and what catastrophes I can expect to encounter while
changing the machine over to new pressures (hence new most every other
setting). Also, it's very hard for me to anticipate the problems I will run
into, as I've never done this before... Give me your worst! Finally, the
cells I will be sorting are very large and very fragile. Does anyone have
tricks for sorting these cells and maintaining viability/integrity?

Thanks so much!
Emily
