B cell assay question

hello,
I am trying to replicate an assay for B cells (CD19+) activated using poke weed mitogen. 
However an unstimulated CD19+ population lies such that quadrant markers need to be moved
around quite a bit.
Although I do see stimulation in activated samples, I would like to know if there is
anything about the behaviour of B cells that I should know about, when performing flow?
