Flowers, I have used just about every stimulus (PMA/iono, Ag, super-Ag, LPS, etc.) to activate cells for intracellular cytokine staining EXCEPT anti-CD3. I would like to activate human T cells with anti-CD3 for intracellular cytokine staining and/or Miltenyi cytokine secretion assay. Question: Does anyone have experience using anti-CD3 activation of human T cells in these assays? Soluble vs. plate bound vs. beads? Particular clones that are better? My major interest is Th2 cytokines, but I'll take any advice. On a similar note, does anyone know of a source for the 64.1 anti-huCD3 hybridoma? I'd love to get my hands on the cells.... Thanks!
