human CCR5 and CCR8

Dear all

does anybody have used with consistent results any monoclonal Abs aginst
human CCR5 and CCR8?

I am using 2D7-APC from pharmingen for CCR5 and clone 191704-APC from
R&D for CCR8.

I am using these antibodies to measure chemokine receptor expression on
anti-CD3 activated human T-cells, either naive or total cells.

First of all nothing can be seen on the resting populations and second,
nothing can be seen on the activated populations.
I also did several tests in which i tested different amount of antibody
(up to 20 µl on 10^5 cells) on activated cells, but i couldn't get any
stain.

I saw from literature that these two antibodies were used in several
papers which used them in systems similar to mine.
They also showed some very good stainings.

The 2D7 datasheet says that staining for chemokine receptors should be
made at room temperature, however the literature reports that staining
was made at 4º C.

I personally stained the same cells for other chemokine receptors such
as CXCR3 and CCR4 at 4º C with excellent and thoroughly consistant results.
Is temperature really an issue?

Can anyone suggest me some sure-shot clones?

Is there any population of cels in the human PBMC-pool that do express
CCR5 and CCR8 so that i can test my antibodies on them?

I thank you very much your kind attention
