Hello, I have a collegue who wants to use DSred2 expression system under CMV promoter. He is going to use DSred2 based vector for retroviral packaging to detect an efficient packaging cell clones. If you have successfully used DSred2 fluorescence system (BD/Clonetech) with either calibur or Aria based FACS analysis, could you please give us the details of what excitations laser as well as emission filters you used? Product insert says that it can be excited with 488nm blue laser, but looking at its excitation peak, it seems unlikely to be shining at 488. What emission filters? I searched the purdue list and there were some suggestions to use PE filters. Is that true, any success? How about 588nm band pass filter that we use for PI? Also, there are some suggestions with respect to peak times after transfection (that it matures late? and that is is gree to start with?), any thoughts on the timings after transfections that you had success with? your experience could save us lot of time standardizing this system, so any input is appreciated.
