Elizabeth,

The simple answer is that there is no reason to do so.

Are you worried about other non-reagent fluorescence spilling into the one 
fluorochrome ala autofluorescence.  There are ways to deal with that.

Could you explain why you would want to apply compensation to a single 
fluorochrome?
