Hi All, I will really appreciate if somebody will help me. I am familiar with conjugation of different dyes including R-PE and APC to mAb. Recently, I started to prepare our own tandem dyes based on R-PE and AlexaFluor647, 680 and 750 dyes. However we found that our tandems undesirably stain human monocytes. So we bought original samples of tandems from Molecular Probes (Invitrogen) just for comparison but they did the same problems. When I compare commercially available antibody conjugated to PC5 (i.e. Immunotech) I had no troubles with non-specific binding to monocytes.
