Dear Flowers I would be grateful if you could kindly send me the protocol of studying activation markers (CD69, CD25, CD71) on CD3 PHA activated peripheral blood mononuclear cells (PBMC). I used BD monoclonal antibodies of CD69, CD25 and CD71 (PE) as well as CD3 FITC for 48 hours PHA (10 microgram/ml) activated PBMC in 24 wells culture plates. The major problem was FSC/ SSC mixed pattern which was changed strangely and dual colors were not separated properly. Please kindly let me know how I can set up the experiment.
