Adam, PBS does not break down at high sorting pressure, and has always worked pretty well as sheath for most of the lymphoid sorts I have been involved with over the years. However, some cells do seem to come through the process much better if the sample buffer has additional buffering capacity, and it is not uncommon to see successful labs use 10 - 25 mM HEPES added to the sample buffer (usually PBS or Hank's) in addition to reasonable amounts of protein. He said that PBS can breakdown with high pressure sorting and that we should use a normalized saline as our sheath fluid instead. Does anyone else use this and can you supply me with the company and the catalog number of the product. Thanks.
