Hi Gayle,

One can use several agents to induce apoptosis in human PBMNC. A simple
method we use that works well is with H2O2,  read out as Annexin V/PI on
flow. You get a nice dose response after culturing freshly isolated
PBMNC for 18 hours in 0.05-10 mM H2O2. The data can be expressed as an
apoptotic index (early apoptotic AnnV+PI- and late apoptotic AnnV+PI+).
Hint: we find best results with H2O 2 bought at our corner drugstore,
don't bother with high grade stuff.
A Disclaimer:  The above terminology does not take into account
complexities about whether or not Annexin V expression distinguishes
apoptosis from necrosis.
