Hello all, I seem to be getting some contradicting info re. propidium iodide staining for live/dead differentition and the ability to do so without having a UV laser. Judging from the excitation spectra, it looks like you can get some excitation with a 488 laser. I am wondering if this is enough to allow for discrimination of live dead cells without having to use an inappropriately high gain. I only have access to a 488 laser and would like to run assays using PI to gate out dead cells and FITC conjugated antibodies for my antigen detection So, does anyone out there run live/dead assays using PI and only a 488 laser?
