Hi Everyone, I am in the early stages of setting up a Cox-1&2 flow assay in human whole blood monocytes for our laboratory. The assay will be used to monitor the in vitro inhibitory effects (if any) of certain extracts on both Cox-1 & 2. I am using LPS to stimulate Cox-2 levels, with unstimulated monocytes going from approx. 3% to 95% Cox-2 positive (post stimulation) However LPS appears to have little if any effect on Cox-1 levels with both pre & post stimulation levels being around the 1% mark. It would therefore be very difficult to assess any inhibitory agent effect on Cox-1 at those levels. Does anyone know of a Cox-1 stimulatory agent that I could incorporate into the assay, so that I can monitor any inhibitory effects on Cox-1 as well as Cox-2 ? Any thoughts or comments would be really appreciated.
