Dear flowers!
Sometimes after ac-lysing the white cells seem to suffer and when doing flow cytometry we
see a bunch of cells from the negative quadrant heading towards upper right. There is
obviously unspecific staining and the cells are probably dead or dying. The phenomenon
may be present when lysing whole blood or bone marrow - most often it happens to one of
the samples from the patient, not with both even when being prepared at the same time.

We have checked pH in the lysing solution and the fixative, carefully checked the lysing
time, tried to vary temperature in the solutions we are using, and tried to avoid using
2% PFA as the last step in the staining procedure.

Anyone else having the problem or the solution to this problem?
