In my opinion the original article in the NEJM had ambiguous flow data and if you look closely the gates are "floating". I'm not surprised. This runs counter to the idea that ZAP-70 expression works as a surrogate marker for unmutated IgVH gene status, which occurs in roughly half of CLL patients. We are running on a Beckman Coulter XL, staining cells after permiabilization with Coulter IntraPrep reagent. Is anyone getting unambiguous results with ZAP-70 testing by flow?
